No estoy familiarizado con PIL, pero sé que es muy fácil poner un montón de imágenes en una cuadrícula en ImageMagick.

¿Cómo puedo, por ejemplo, poner 16 imágenes en una cuadrícula 4 × 4 donde puedo especificar el espacio entre las filas y las columnas?

Esto también es fácil de hacer en PIL. Cree una imagen vacía y simplemente pegue las imágenes que desee en cualquier posición que necesite usando paste. Aquí hay un ejemplo rápido:
